Description: Affinity capture with low flow chromatography and high-resolution mass spectrometry (AC-LCMS) is a hybrid method for bioanalysis of pharmacodynamic markers and protein/peptide therapeutics. It achieves low LLOQs, accuracy, precision, broad linear dynamic range, and robustness. We present two case studies: 1) TL1A biomarker analysis in human serum with LLOQ of 40 pg/mL using hybrid AC-LCMS, and 2) TMT chemistry for bioanalysis of known mAb therapeutics spiked in serum.

Learning Objectives:

  • AC-MS hybrid methodology with Nanoflow/microflow chromatography combined with HRMS have demonstrated to achieve appreciable LLOQs, with several orders of magnitude broad linear dynamic range (LDR) with robustness, manageable carry over.
  • Achieve ultra-high sensitivity as low as 40pg/mL LLOQ for serum biomarkers like TL1A using affinity capture low flow chromatography coupled to high resolution mass spectrometry.
  • Applicability evaluation of the TMT based hybrid workflow for quantitative bioanalysis of mAbs in serum by analyzing qualification parameters employing a 96-well based sample preparation approach.
