Description: Choice of extraction approach is fundamental to successful qPCR/dPCR bioanalytical studies. We performed multiple controlled studies comprising thousands of samples evaluating reagent variability, automated extraction approach, multiple PCR targets (RNA and DNA) and cell-heavy and cell-free matrices. These runs included evaluation of Recovery, False Positivity, Precision and Accuracy. Extraction approach and matrix primarily influenced outcomes. In addition, ways to address inter-run variability will be discussed.
