Purpose: Compare high-throughput RNA extraction kits to improve downstream reliability.

Methods: Samples (50 mg) were homogenized via bead-beating. RNA extraction followed the manufacturer's kit protocol, with Xeno IPC spiking, on the KingFisher Flex platform in 8 replicates. RNA purity and yield were measured by Nanodrop spectrometer. Extraction efficiency (EE) was evaluated via RT-qPCR.

Results: Figure 1 shows RNA purity, Figure 2 displays RNA yield for the extraction kits. Table 1 outlines their EE. High RNA purity is consistently observed, though RNA yield varies. EE varies among kits decreasing across brain, liver, spleen, heart, kidney, and lung tissues.

Conclusion: Studied RNA extraction kits provide high purity but vary in yield and EE indicating kit selection and method optimization is crucial for downstream applications.
