Nano PharmaSolutions (NPS) has developed a novel drug nanosizing technology by evaporating solid molecular drugs into gas-phase, which is then nanocoated onto a hydrophilic excipient such as mannitol under ultra high vacuum. This binary nanoformulation has improved solubility of BCS Class II and IV drugs and better bioavailability. One example of this nanoformulation is nasal spray form of apomorphine freebase (APO-FB) which is only available in the HCl salt (APO-HCl) injection form due to the insolubility of the freebase. Comparison of the canine PK data of APO-FB nasal delivery against subcutaneous injection of APO-HCl showed faster absorption of APO-FB as nanoformulation, which leads to faster time of "ON" for Parkinson's Patients than the subcutaneous injection formulation. The duration of drug absorption is possible through varying particle size distribution of the nanosized APO-FB. Powder nasal delivery is attractive for CNS diseases for fast delivery, better stability, and potential nose-to-brain delivery.
