We investigate in vivo stability and biotransformation of multiple antibody-drug conjugates (ADCs) in the intact form using high-resolution mass spectrometry. In comparison with other approaches, HRMS enables the comprehensive identification of ADC biotransformation products with more confidence. It is particularly beneficial for characterizing ADC modifications with small mass changes such as partial drug loss and hydrolysis. This strategy has significantly enhanced our capability in elucidating ADC biotransformations and better understanding ADC efficacy and safety in vivo.
Learning Objectives:
Upon completion, participants will be able to understand the key elements in the intact mass analysis of ADC and its biotransformation products using HRMS.
Details on sample preparation, including immunoaffinity capture, will be discussed to demonstrate the importance of analyte cleanup and enrichment.
